I've just tried this exactly command line on OSX on 10.1 - worked as expected, TokuDB was not built.

I used -DWITHOUT_TOKUDB=1 as suggested by Kristian, and that built OK, but -DPLUGIN_TOKUDB=NO doesn't seem to be doing the trick.
Yes, old WITH_xxx/WITHOUT_xxx variables are still supported. They're internally converted to PLUGIN_xxx values. But PLUGIN_xxx should work on their own too.
I have cmake 3.0.1, in case that's relevant.
